In 1957, the Sqn was heavily involved in internal security duties at RAF Akrotiri as part of the overall campaign against Ethniki Organosis Kypriakon Agoniston (EOKA) terrorists. Due to the rising tension in the Middle East, the Sqn was deployed operationally in the Light Anti-Aircraft role and in Dec 1958 the Sqn received the first batch of its new Bofors L70 guns. The Sqn saw continued service in the Internal Security role in an around Nicosia until, due to the worsening situation in the British colony, it was deployed to Aden in 1965. The Sqn returned to RAF Akrotiri toward the end of the year and was given notice of its return to the UK having completed 15 years uninterrupted service in the Middle East.

By Sep 1966, the Sqn had completed its move to the UK and was integrated into Transport Command. However, on Aug 1967 the Sqn once again returned to RAF Akrotiri as part of No 3 Wing together with 34 Sqn RAF Regt. Between 1968 and 1974 the Sqn undertook a number of tasks as part of the Immediate Reaction force including both Internal Security and Light Anti-Aircraft duties.

In Jan 1975 the Sqn redeployed to RAF North Luffenham to convert to the new Rapier missile system. After successful conversion, the Sqn took up residence at RAF Leuchars as part of No 11 Gp and in Aug 1977 Her Majesty the Queen approved the award of a Standard to 27 Sqn RAF Regt for 25 years service. In 1979 the Sqn became the first unit within the British Forces to be declared fully operational with the DN 181 Blindfire Rapier system. Between 1980 and 1982 the Sqn shared the roulement commitment of providing Short Range Air Defence at Belize International Airport with No 48 Sqn RAF Regt.

In 1995, the Sqn moved to RAF Scampton as a cadrised Sqn within the RAuxAF Regt but in Sep 1998, after continuous service since 1951, the On 1st Apr 1999, No 27 Sqn and 1 Royal Tank Regt merged to form the Joint Nuclear, Biological and Chemical Regt (Jt NBC Regt), the first truly dual-service front-line unit. No 27 Sqn is now responsible for the manning and management of the Biological Detection Capability within the Jt NBC Regt.
